摘要
目的:建立测定变通丸中盐酸小檗碱含量的方法。方法:采用反相高效液相色谱法。色谱柱为SunFireTMC18(250mm×4.6mmi.d.,5μm),流动相为1.5mmo·lL-1十二烷基硫酸钠溶液(以磷酸调pH5.0)-乙腈(梯度洗脱),流速为0.8mL·min-1,柱温为25℃,检测波长为265nm。结果:盐酸小檗碱的质量浓度在60.8~304.0μg·mL-1范围内与峰面积积分值呈良好线性关系;平均加样回收率为97.59%,RSD=1.34%(n=6)。结论:本法简便、快速、重复性好,结果准确、可靠,可作为变通丸中盐酸小檗碱含量测定的方法。
OBJECTIVE:To establish the method for the content determination of berberine hydrochloride in Biantong pills.METHODS:RP-HPLC method was adopted.The determination was performed on SunFireTM C18 column(250 mm×4.6 mm,5 μm) with mobile phase consisted of 1.5 mmol·L-1 sodium dodecyl sulfate(pH value adjusted to 5.0 with phosphoric acid)-acetonitrile(gradient elution) at flow rate of 0.8 mL·min-1.The column temperature was 25 ℃ and detection wavelength was set at 265 nm.RESULTS:The linear range of berberine hydrochloride was 60.8~304.0 μg·mL-1 with an average recovery of 97.59%(RSD=1.34%,n=6).CONCLUSION:This method is simple,rapid,reproducible,accurate and reliable for the content determination of berberine hydrochloride in Biantong pills.
