摘要
目的:建立体外胚胎和子宫内膜共培养体系,观察不同浓度的IL-1β和瘦素(leptin)对ICAM-1、MMP9的影响,进而探讨IL-1β和leptin对胚胎早期着床的调节作用。方法:采用免疫细胞化学法检测子宫内膜中ICAM-1和MMP9的表达;采用ELISA方法分别检测经IL-1β、leptin和IL-1ra干预共培养模型后培养液中ICAM-1和MMP9的表达。结果:免疫组化结果显示,ICAM-1主要在腺体细胞中表达,基质细胞仅有少量的表达,而MMP9在子宫内膜基质细胞和腺体细胞中均有表达;ELISA结果显示,与对照组相比,ICAM-1分别经10 ng/ml的IL-1β,100 ng/ml的leptin干预后表达上调(P<0.05),而经0.1ng/ml、1ng/ml的IL-1β,1 ng/ml、10 ng/ml的leptin,0.25μg/ml、2.5μg/ml、25μg/ml的IL-1ra干预后表达无明显变化(P>0.05);MMP9分别经1 ng/ml、10 ng/ml的IL-1β,1 ng/ml、10 ng/ml、100 ng/ml的leptin干预后表达上调(P<0.05),而经0.1 ng/ml的IL-1β,0.25μg/ml、2.5μg/ml、25μg/ml的IL-1ra干预后表达无明显变化(P>0.05)。结论:一定量的IL-1β和leptin可以增加共培养模型中ICAM-1和MMP9的表达。
Objective:To study the mechanism of interleukin-1β(IL-1β) and leptin on embryo implantation in the early stage by observing the expression of intercellular adhesion molecule-1(ICAM-1) and metalloproteinase 9(MMP-9) intervened by IL-1β or leptin which were closely related to the implantation in the vial model of embryo and endometrium.Methods:The expressions of ICAM-1 and MMP-9 in the endometrium were detected by immunohistochemistry.However,the expressions of ICAM-1 and MMP-9 in the medium of model which have been intervened seperately by IL-1β,leptin or interleukin-1 receptor antagonist(IL-1ra) were detected by ELISA..Results:Immunohistochemistry:ICAM-1 was mainly expressed in the glandular cells,rarely in stromal cells,but MMP-9 was expressed in endometrial stromal cells and glandular cells.The result of ELISA showed that the expression of ICAM-1 in the medium was upregulated after intervened by IL-1β(10 ng/ml) or leptin(100 ng/ml)(P0.05).But after separately intervened by IL-1β(0.1 ng/ml,1 ng/ml),leptin(1,10 ng/ml) or IL-1ra(0.25,2.5,25 μg/ml),they had no significant difference compared with control group(P0.05).The result of ELISA showed that the expression of MMP-9 in the medium was upregulated after intervened by IL-1β(1,10 ng/ml) or leptin(1,10,100 ng/ml)(P0.05).But after separately intervened by IL-1β(0.1 ng/ml) or IL-1ra(0.25,2.5,25 μg/ml),they had no significant difference compared with control group(P0.05).Conclusion:Certain amounts of IL-1β and leptin can upregulate the expression of ICAM-1 and MMP-9.
