摘要
目的探索一种简易的同时测定人血浆丙泊酚和利多卡因的血药浓度分析方法,为临床研究两种药物之间的相互作用提供科学依据。方法用环己烷提取血浆中的丙泊酚和利多卡因,用高效液相色谱法(high performance liquid chromatography,HPLC)测定丙泊酚和利多卡因的含量。以Shim-packVP-ODS为分离柱,乙腈∶甲醇∶水(10∶60∶30)为流动相,流速为1 mL.min-1,采用外标法,用紫外检测,检测波长:1~6 min用220 nm,6~15 min用270 nm。结果丙泊酚和利多卡因的血药浓度与峰面积比在0.5~32μg.mL-1的范围内呈线性关系(r=0.9994,r=0.9986),检测限分别为0.05μg.mL-1和0.1μg.mL-1,日内和日间偏差小于5%(n=5,n=6)。结论本方法简便,具有较高的精密度和准确度,适用于临床丙泊酚和利多卡因两种药物血药浓度的同时检测。
Objective To develop a method for the determination of propofol and lidocaine concentration in plasma by high performance liquid chromatography(HPLC) and to provide scientific analysis for studies of the pharmacology of propofol and lidocaine in clinical anesthesia.Methods The propofol and lidocaine in plasma was extracted with cyclohexane and was determined by HPLC.The propofol and lidocaine was separated on Shim-pack VP-ODS column with a mobile phase of acetonitril∶ methanol∶ water(10∶ 60∶ 30) at a flow rate of 1 mL·min-1 and detected at 220 nm during 1~6 min and at 270 nm during 6~15 min.Results The linearity between concentrations and peak area ratio was obtained from 0.5 μg·mL-1 to 32 μg·mL-1(r=0.9994,r=0.9986).The detection limits were 0.05 μg·mL-1 for propofol and 0.1 μg·mL-1 for lidocaine.The within-day and between-day variation coefficients were less than 5%(n=5,n=6).Conclusion The reversed-phase HPLC is precise and can be used to measue propofol and lidocaine plasma levels in clinic.
