摘要
为探明人参细胞悬浮过程中植物生长调节剂对细胞生长和皂苷合成的影响,调查IBA、NAA、2,4-D单独使用及IBA与BA和IBA与KT混合使用的效果,结果表明,在2,4-D处理中细胞生物量虽然高于IBA和NAA,但总皂苷含量低于IBA和NAA处理;在生长素单独使用处理中,IBA 7 mg/L处理的皂苷生产量最高(72.9 mg/L).当IBA 7 mg/L与BA或KT混合使用时,促进细胞生长和皂苷合成,其中IBA 7 mg/L+BA0.5 mg/L和IBA 7 mg/L+KT 0.5 mg/L处理中,皂苷生产量明显提高,分别达到115.92和120.98 mg/L.因此,在进行人参细胞悬浮培养时,IBA 7 mg/L与BA 0.5 mg/L或KT混用可高效生产人参皂苷,为食品及药品生产提供安全的原料.
To select appropriate plant growth regulators during cell suspension culture of Panax ginseng, the effect of IBA, NAA, 2,4-D, and combination of IBA with BA or KT on cell growth and ginsenoside production were investigated. The results showed that the better cell growth was observed in MS medium supplemented with 1. 0 mg/L 2,4-D while higher ginsenoside contents were analyzed in IBA and NAA treatments, the maximum ginsenoside productivity (72.9 rag/L) was found in 7 mg/L IBA. In addition, the combination of IBA with different concentrations of BA or KT promoted cell biomass and ginsenoside contents~ MS medium supplemented with 7 mg/L IBA and 0.5 mg/L BA or 0.5 mg/L KT was optimal, more than 115 mg/L of ginsenoside productivity was obtained. Our finding indicates that 2,4-D can be replaced by IBA during cell suspension culture of Panax ginseng, which brings not only high ginsenoside productivity but also safe productions in the industry of medicine and food.
