摘要
目的建立测定人血浆中地西泮和咪达唑仑质量浓度的高效液相色谱法。方法色谱柱采用Waters C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(55∶45),柱温30℃,流速1.0 mL/min,检测波长220 nm。结果地西泮和咪达唑仑的最低检测质量浓度为5 ng/mL,进样质量浓度的线性范围为0.05~10.0μg/mL(r>0.990 0);咪达唑仑和地西泮日内精密度的RSD分别为3.09%~5.33%和1.56%~4.25%,日间精密度的RSD分别为4.72%~6.52%和2.59%~6.22%,方法回收率分别为94.02%~97.19%和96.07%~105.64%。结论所用方法简便、快速、准确,适用于地西泮和咪达唑仑体内血药浓度和生物等效性的测定。
Objective To determine diazepam and midazolam in human plasma by HPLC.Methods The HPLC separation was carried out on a Waters C18 column(250 mm×4.6 mm,5 μm),using a UV detector and the wavelength was 220 nm,and mobile phase was acetonitrile-water(55∶45).Results For diazepam and midazolam,the limits of detection were 5 ng/mL.The concentration of midazolam and diazepam showed a linear plot in the range of 0.05-10.0 μg/mL(r 0.990 0).Within-day and between-day precisions expressed as the relative standard deviation(RSD) for the method were 3.09%-5.33%,1.56%-4.25%,4.72%-6.52%,and 2.59%-6.22%,respectively.The extraction recoveries were 94.02%-97.19%and 96.07%-105.64%,respectively.Conclusion The method is proved to be accurate,sensitive and reliable.It is suitable for the simultaneous determination in human plasma of diazepam and midazolam.
