摘要
采用硫酸铵法、辛酸二步法、DEAE-Sephadex A50 离子交换柱层析法,从杂交瘤小鼠腹水中纯化人型结核杆菌H37Rv 株特异性抗原TB4 的单克隆抗体,从回收率、纯度和活性3 个方面进行比较。结果显示,硫酸铵法回收率最高,层析法纯度最高,3 种方法对单抗的活性均无大的影响。认为:辛酸二步法可有效地去除白蛋白,适用于大量腹水中单抗的提取、纯化,具有简单、省时等优点;
Ammonium sulfate,caprylic acid precipitation and ion exchange chromatographical methods have been used to purify monoclonal antibody(McAb) against TB4 antigen of Mycobacterium tuberculosis H 37 Rv strain from ascitic fluid in Balb/c mice.The results showed that ammonium sulfate precipitation method could have the highest harvest rate and ion exchange chromatography method could have the higest purity.These methods of purifying McAb could not impair the antibody activety.Caprylic acid method can remove albumin effectively and the purification of anti TB4 McAb in ascitic fluid is simple and easy to perform,less time comsuming and less cost.Ion exchange chromatographical method is applicable to analysis.
