摘要
以变叶金银木为外植体,探讨了不同生长调节物质对外植体分化、增殖和生根的影响。结论如下:(1)变叶金银木组织培养最适宜的外植体是半木质化带芽枝段,以70%CH2(OH)560 s+0.1%HgCl2消毒4~5 min为最好;(2)在不添加任何激素的MS培养基中20 d后能诱导出幼芽,适宜的增殖培养基为MS+6-BA 0.3 mg.L-1+NAA 0.1 mg.L-1,生根培养基为1/2 MS+IBA 0.2 mg.L-1;(3)生根试管苗适宜的开瓶锻炼时间为3 d,选择直径为0.5~1 mm的试管苗移栽到蛭石培养基上能够获得76.7%~86.6%的成活率。
Using Lonicera maackii'Bian ye 'as explants,influences of different plant growth hormones on differentiation,multiplication and rooting were examined.The results show that(1)the optimal explant material was tender stem with single bud,sterilized 1 minute in 70% CH2(OH)5 and 4~5 minutes in 0.1% HgCl2;(2)the young buds cound be induced after 20 days cultured on the medium MS,the optimal subculture and rooting medium were MS+6-BA 0.3 mg·L-1+NAA 0.1 mg·L-1and 1/2 MS+IBA 0.2 mg·L-1 respectively;(3)the glass seedling rooting condign open the glass bottle hardened times was 3 days,than the diameter 0.5~1 mm glass seedling was transplanted on vermiculite,the survive rates of glass seedling were 76.7%~86.6%.
