摘要
目的介绍一种移植预处理后肝脏内皮的电镜制作方法。方法将30只C57BL/6小鼠随机均分为A组(60Co 7.5 Gy致死量照射)、B组(60Co 5.0 Gy减低剂量照射)和C组(正常对照),通过透射电镜定期观察肝脏内皮变化。结果透射电镜下可见:A组肝脏内皮细胞核碎裂、内皮破坏严重;B组染色质边集,内皮无明显损伤;C组未见明显异常。结论对肝脏组织进行恰当的处理后,透射电镜下能清楚地观察到内皮细胞表面结构和内壁损伤的形态改变。
Objective To introduce a method of making electron microscope of the liver endothelium after the transplantation conditioning.Methods Thirty C57BL/6 mice were randomly and equally divided into three groups of A(exposed to lethal irradiation with 60Co 7.5Gy),B(exposed to reduced irradiation with 60Co 5.0Gy) and C(normal controls).After irradiation,the changes of liver endothelium were regularly observed by transmission electron microscope.Results Under the transmission electron microscope,cell karyorrhexis of the liver endothelium and badly injured endothelium were found in group A,while the chromatin margination and obsoletely injured endothelium were observed in group B.And there was not obvious abnormality found in group C.Conclusion The surface structure and intine injury changes of liver endothelium that have been properly treated can be clearly observed by the transmission electron microscope.
