摘要
为了解运动后大鼠肌细胞IRS-2、GLUT4蛋白含量及IRS-2磷酸化的动态变化及时间特征。制备II型糖尿病大鼠模型。将SD大鼠分为正常对照组、糖尿病对照组和糖尿病运动组。用Western blot免疫印迹法检测运动后IRS-2、GLUT4蛋白含量及IRS-2磷酸化程度。结果得出:与糖尿病对照组相比,运动后1 h、3 h组、6 h、12 h、24 h和48 h股四头肌IRS-2蛋白含量分别升高6.8%、23.7%(P<0.01)、20.7%(P<0.01)、10%、5.4%和2.7%;GLUT4蛋白含量分别显著升高17.3%、32.7%、42.3%、46.2%、51.9%和45%(P<0.01);IRS-2磷酸化分别升高49%、51.4%、45.1%、43.1%、21.6%(P<0.01)和9.8%(P<0.05)。结论:一次性中等强度的运动对IRS-2、GLUT4含量和IRS-2磷酸化产生显著的影响,其中GLUT4含量和IRS-2磷酸化升高的效应可持续到运动结束的48 h。
Objective:To explore the dynamic effects of exercise on protein content of IRS-2, GLUT4 and IRS-2 tyrosine phosphorylation in DM rats. Method: SD rats were divided into normal control group (NC),diabetes mellitus group (DMC) and exercise group. The exercise group did one-bout exercise for 50min. The protein contents of IRS-2, GLUT4 and IRS-2 tyrosine phosphorylation were measured using Western bolt analysis. Result:The contents of IRS-2 protein in 1 h, 3 h, 6 h, 12 h, 24 h and 48 h groups increased up to 6.8 %,23.7 % (P〈0. 01), 20. 7 % (P〈0. 01), 10 %,5.4 % and 2.7 % ; GLUT4 protein in 1 h,3 h,6 h,12 h,24 h and 48 h groups increased up to 17. 3%,32. 7%,42. 3%,46. 2%,51.9% and 45% (P〈0.01). The tyrosine phosphorylation of IRS-2 protein in 1 h, 3 h, 6 h, 12 h, 24 h and 48 h groups increased up to 49%,51.4%,45. 1%0,43. 1%,21.6%0 (P〈0. 01) and 9.8G(P%0.05) respectively compare with the DMC group. Conclusion: The results showed that the contents and the tyrosine phosphorylation of IRS-2 and GLUT4 protein have the obviously effect after once bout exercise,and the effect could last for 48 h after exercise.
