NSE、GFAP及Notch1在大黄三七散改善急性脑出血大鼠神经功能缺损中的表达及其意义 被引量:5

Expression of NSE,GFAP and Notch1 and Their Significance on the Improvement of Duanghuasanqi Powder on Neural Function Deficiency in Rats with Intracerebral Hemorrhage

摘要 研究神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)及Notch1受体蛋白在中药大黄三七散改善急性脑出血(ICH)大鼠神经功能缺损中的表达及其意义。以肝素胶原酶Ⅳ法制作大鼠脑出血模型,考察了施药前后ICH大鼠神经功能评分、脑系数及脑含水量、NSE、GFAP及Notch1在mRNA及蛋白水平的表达量及相关性。结果表明,大黄三七散可明显改善ICH大鼠神经功能缺失症状,显著降低脑系数并减少脑含水量,其对ICH大鼠血清内的NSE、GFAP及Notch1蛋白表达量的升高均有不同程度的抑制作用。ICH大鼠脑血清中NSE的表达与Notch1在mRNA及蛋白水平均呈极显著正相关,而与GFAP的表达在mRNA及蛋白水平均无相关性。而GFAP表达与Notch1正相关。NSE、GFAP及Notch1蛋白的关联性表达研究,表明三者不仅可作为判断急性脑出血损伤发展程度的重要生物标记,还可作为联合选择拮抗蛋白表达的共同药物作用靶点,对急性脑出血的防治具有重要意义。 This paper is to study the expression of NSE,GFAP and Notch1 and their significance on improvement of Duanghuasanqi powder on neural function deficiency in rats with intracerebral hemorrhage.The ICH mouse model was established by the collagen enzymes IV infusion method.Neurologieal scale,cerebral indexes,water content,the the expression of NSE、GFAP and Notch1 and their correlation were analyzed.The results showed that adminstration of the Duanghuasanqi powder can obviously imporve the neurologieal scale,decreased the cerebral indexes and water contents and the induced expression of the NSE,GFAP and Notch1 proteins.The expression of NSE and Notch1 demostrated significantly positive correlative,but has no correlation with GFAP.Whereas,the the expression of GFAP and Notch1 also showed positive correlative.In conclustion,we indicated that the correlative expression of the NSE,GFAP and Notch1 proteins can not only used as an essential marker for the judgement of the development degree of ICH damage,but also can be used as an action target for drug-candidate screening for the sure of the prevention and cure of ICH.
