NF—KB和VEGF在高氧暴露早产鼠视网膜的表达及地塞米松的干预效应

The interventional effect of dexamethasone on the expression of NF-Kb and VEGF of retina in premature rat mottle with hyperoxia--induced retinopathy

摘要 目的:探讨核因子-KB(nuclear factor—kB,NF—KB)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在高氧暴露早产鼠视网膜的表达规律及地塞米松对NF—KB和VEGF表达的影响。方法:标准SD大鼠的3日龄早产鼠36只随机分为3组,高氧组、干预组、空气组,其中高氧组和干预组均吸入氧气(氧浓度≥90%)7天返回空气5天,空气组一直置同一室内空气中,干预组同时每天腹腔注射地塞米松1.0mg/kg,高氧组和空气组每天腹腔注射等量生理盐水。用免疫组织化学法检测各组早产鼠视网膜NF—KB和VEGF表达水平,记教突破视网膜内界膜的内皮细胞棱。结果:空气组NF-KB、VEGF均呈低表迭,未见或罕见突破内界膜的内皮细胞核;与空气组相比,高氧组突破内界膜的内皮细胞核明显增多,NF—KB及VEGF的表达均显著增加(P分刺〈0.001,0.001,P〈0.01);与高氧组相比,干预组突破内界膜的内皮细胞核明显减少,NF—KB及VEGF的表达均显著下降(P分别〈O.05,0.001,0.05);NF—KB与VEGF,NF-KB与新生血管数及VEGF与新生血管数呈正相关(r分别为0.47,0.56,0.45,P分别〈0.05,0.01,0.05。结论:高氧暴露旱产鼠视网膜内可观察到VEGF和NF—KB的高表达,地塞米松使高氧暴露早产鼠视网膜NF--KB和VEGF的表达明显下降,突破视网膜内界膜的内皮细胞核数量明显减少,减少新生血管的形成,对高氧性视网膜痛有一定的保护作用。 Objective:To investigate the effects of dexamethasone on the expression of NF--Kb and VEGF in premature rat modle with hyperoxia --induced retinopathy. Methods:Thirty--six premature Sprague--Dawley rats were randomly divided into into 3 groups, hyperxia group,dexamethasone treatment group and air group, hyperxia group and dexamethasone treatment group were exposed to ≥95% 02 for 7 days,then exposed to air for 5 days, air group exposed to room air. Dexamethasone treatment group were intraperitoneal injeetied of dexamethasone 1.0mg/kg,hyperxia group and air group were intraperitoneal injecticd same dose of 0.9% sodium chloride. Expressions of hypoxia inducible factors-1α(HIF-1α)and vascular endothehal growth faetor(VEGF)were examined with immunohistoehemieal staining, The numbers of endothelioeyte nuclei of neovascularization which broke through the retinal inner limiting memberane were recorded. Results In air group, the numbers of endotheliocyte nuclei of neovaseularization which broke through the retinal inner limiting memberane were not found or seldem found,expressions of HIF-1α and VEGF were low level;while compared with the air group, all the three above of hyperxia group all greatly increased,p〈0. 001,0. 001,0.01 ,respectively. Compared with the hyperoxia group,all the three above of dexamethasone treatment group all greatly decreased, p〈 0. 05,0. 001,0. 05, respectively. There are positive correlation between NF- KB and VEGF, NF- KB and the numbers of endothdioeyte nuclei of neovaseularization which broke through the retinal inner limiting memberane, VEGF and the numbers of endotheliocyte nuclei of neovaseularization which broke through the retinal inner limiting memberane, r= 0. 47 , 0. 56,0. 45, P〈0. 05,0. 01,0. 05, respectively. Conclusion Dexamethasone have protective effects in hyperoxia--induced retinopathy in premature rat.
