摘要
目的探讨不同浓度的羟乙基淀粉类血浆代用品(贺斯6%HAES200/0.5)对正常人体免疫系统白介素-10(IL-10)和白介素-12(IL-12)的影响,以检验其是否抑制正常人体的免疫系统。方法正常志愿者30名,严格无菌条件下抽取静脉血各20ml,并以肝素抗凝,在超净台内分离出单个核细胞,计数后将细胞平均分成5组:1640培养液阴性对照组、10%血浆对照组、10%贺斯试验组、20%贺斯试验组、40%贺斯试验组,然后再将各组分成阴性对照组、加脂多糖(LPS)、加植物血凝素(PHA),使单个核细胞在不同的培养液中培养48h后,离心提取上清液,以酶联免疫吸附法(ELISA法)检测各上清液中细胞因子IL-10、IL-12的浓度。结果 3个浓度的贺斯试验组与1640培养液及血浆对照组在3种环境(阴性对照、加LPS、加PHA)中比较,虽然3种细胞因子的浓度值稍有参差,但差异无统计学意义(P>0.05);各组阴性对照、LPS及PHA间两比较差异均有统计学意义(P<0.05)。结论贺斯在体外试验中不抑制正常人体免疫系统机能。
Objective To observe the influence of colloid plasma substitutes(hydroxyethyl starch) with different concentrations on IL-10 and IL-12 in healthy subjects,and to evaluate weather hydroxyethyl starch(HS) could inhibit the immune system of healthy people.Methods 30 healthy volunteers were enrolled in the investigation whose venous blood 20ml was taken out under sterile condition,and the blood samples were anticoagulated by heparin,then the concentrations of IL-10 and IL-12 were detected by ELISA in five groups:1640 culture medium control group,10% plasma group,10% HS group,20% HS group and 40% HS group.Results As compared with those in 1640 culture medium control group and 10% plasma group,although the levels of IL-10 and IL-12 in three trial groups of different concentrations HS had little difference under three conditions(negative control,plus LPS,plus PHA),there were no significant differences among them(P0.05),however,there were significant differences between the two groups,respectively among negative control groups,LPS groups and PHA groups(P0.05).Conclusion The hydroxyethyl starch does not inhibit immune system of healthy persons in vitro.
