摘要
目的 探讨血管内皮生长因子(VEGF)在实体肿瘤中的作用。方法 应用分子克隆技术构建人全长VEGFcDNA反义基因真核表达载体,并用脂质体法转入人肝癌细胞系(HepG2) ,观察转染后肝癌细胞在裸鼠体内的生长及瘤体内微血管的通透性。结果 各组细胞在裸鼠体内的成瘤时间无明显差别,但反义核酸转染组肿瘤的生长速度及瘤体的重量均明显落后于其他组,肿瘤内血管的通透性也显著降低。结论 VEGF在实体肿瘤的生长过程中起着十分重要的作用。
Objective To explore the role of vascular endothelial growth factor (VEGF) in solid tumor. Methods The VEGF antisense gene eukaron expression vector was constructed with molecular cloning, and transfected into hepatocellular carcinoma cell line (HepG2) by lipofectin technique. The growth of transfected cell line in nude mice and the vascular permeability in tumor were examined. Results\ Though the time for oncogenesis in nude mice was not different, the growth rate, the weight of the tumor and the permeability in tumor were markedly lower in transfected group than those in other groups. Conclusion\ VEGF might play an important role in the growth of the solid tumor; Anti vascular is a new method in the treatment of the solid tumor.
