骨髓间充质干细胞抑制Wistar大鼠脊髓损伤区神经元凋亡的实验研究 被引量:4

摘要 目的探讨骨髓间充质干细胞(BMSCs)是否具有抑制神经元凋亡的作用。方法 Wistar大鼠120只随机平均分为4组:对照1组为空白对照,对照2组、两治疗组均采用改良Allen法建立大鼠下胸段脊髓损伤模型。治疗1组、治疗2组分别经尾静脉和脊髓损伤区注入BMSCs。术后用免疫组化检测各组Caspase-3、Bax、Bcl-2的表达。结果术后7天,治疗2组bax、caspase-3阳性细胞表达少于对照2组(P<0.05),治疗1组Bcl-2阳性细胞表达多于对照2组(P<0.05);移植后15天,治疗1组bax以及治疗2组caspase-3阳性细胞表达少于对照2组(P<0.05),治疗1组及治疗2组Bcl-2阳性细胞表达多于对照2组(P<0.05);移植后30天,治疗1组bax阳性细胞表达少于对照2组(P<0.05),治疗1组、治疗2组Bcl-2阳性细胞表达多于对照2组(P<0.05)。结论 BMSCs移植上调Bcl-2蛋白表达,下调Caspase-3、Bax蛋白表达,具有抗凋亡作用,可减轻脊髓继发性损伤。 Objective To approach bone mesenchymal stem cells(BMSCs) whether or not would inhibit the Neuronal apoptosis.Methods To divided 120 Wistar rats into 4 groups randomly.The control group 1 is a blank control,the control group 2 and two treatment groups was produced to spinal cord injury model by modified Allen's crack method.Two treatment groups' BMSCs which were isolated culture in vitro inside the model through the vena caudalis and the partial injury randomly.After transplantation,use the method of immunohistochemical staining detect each groups' Caspase-3,Bax,Bcl-2 expression.Results At the 7th day after transplantation,The Bxa,caspase-3 positive cells expressed in the treatment group 2 less than the control group 2(P0.05),The Bcl-2 positive cells of the treatment group 1 expressed more than the control group2(P0.05);At the 15th day after transplantation,the Bxa positive cells of the treatment group 1 and the caspase-3 positive cells of the treatment group 2 expressed less than the control group 2(P0.05),the Bcl-2 positive cells of the treatment group 1 andthe treatment group 2 expressed more than the control group 2(P0.05);At the 30th day after transplantation,the Bxa positive cells of the treatment group 1 expressed less than the control group 2(P 0.05),the Bcl-2 positive cells of the treatment group 1 and the treatment group 2 expressed more than the surgery group(P0.05).Conclusion BMSCs transplantation could increase the Bcl-2 protein's expression,reduce Caspase-3 and Bax protein's expression,have the role of anti-apoptosis,which could reduce spinal cord secondary injury.
