摘要
目的:研究拳参提取物在大鼠视网膜缺血再灌注损伤时抗氧化作用的影响。方法:取SD大鼠随机分成4组。颈总动脉夹闭法制作视网膜缺血再灌注模型。颈总动脉夹闭1 h再灌注1 h后,行视网膜电流图(ERG)检查;测定血清MDA、GSH-PX及SOD活性。结果:与模型组比较,拳参提取物显著升高ERG幅度、降低MDA浓度、升高GSH-PX、SOD活性。结论:拳参提取物可通过提高SOD及GSH-PX的活性,降低MDA浓度,从而提高视网膜缺血再灌注损伤时的抗氧化作用。
Objective:To study the effect of Polygonum Bistorta L.extract on anti-oxidation and retinal ischemia/reperfusion(RI/R) injury.Methods:Twenty-four male SD rats were divided into sham group,model group of RI/R injury.After clamped CCA for 1 hour and reperfused for 1 hour,the electroretinogram(ERG) was tested;then the vein blood was collected for detecting the contents of MDA in serum,the activity of SOD、GSH-PX by colorimetric method.Results:The amplitude of ERG after the induction of the ischemia-reperfusion injury is significant lower than the forth,and it is markedly enhanced in the Polygonum Bistorta L.extract group.The content of MDA in serum is obviously increased in model group compared with sham group,and that in low-dosage Polygonum Bistorta L.extract is decreased in comparison with model group.The activity of SOD model group are both significantly lower than that of sham group,but those in low-dosage group are higher than that of model group.The content of serum GSH-PX in model group is lower than in sham group,and that in model group is also lower than in low-dosage Polygonum Bistorta L.extract group.Conclusion:Polygonum Bistorta L.extract can protect retina against cellular damage in RI/R injury eye through elevating activity of SOD、GSH-PX and increasing anti-oxidation of retina.
