摘要
目的:探讨五味子乙素(SchB)与5-氟尿嘧啶(5-FU)体外协同作用及对人胃癌细胞增殖及凋亡的影响。方法:MTT法检测不同浓度SchB单用或与5-FU联用对细胞增殖的抑制作用;光学显微镜观察细胞生长状态变化并计数细胞分裂指数;电镜观察药物对细胞超微结构的影响;TUNEL法检测细胞凋亡。结果:SchB单独应用对细胞增殖的抑制作用弱,与5-FU联用抑制作用显著,呈剂量依赖性;药物处理48h后,联合用药组细胞分裂指数为(1.1±0.28)%,与SchB组(13.7±0.43)%、5-FU组(5.1±0.52)%比较显著降低,P<0.05;电镜下可见细胞皱缩,胞膜界限模糊,核膜消失,核质浓缩及凋亡小体形成,以双药联用最为显著。TUNEL法分析示,联合用药组细胞凋亡率为(73.9±5.1)%,与SchB组(16.4±2.4)%、5-FU组(34.6±3.2)%比较差异有统计学意义,P<0.05。结论:SchB与5-FU联合应用能有效抑制胃癌细胞增殖,促进细胞凋亡,二药具有协同增效作用。
OBJECTIVE:To investigate the effect of combination sehisandrin B (Seh 13) with 5-fluorouracil (5 FU) on the proliferation and apoptosis of human gastric cancer cells and the synergy between them. METHODS: The growth inhibitory effects of Sch B and 5-FU on cells were studied by MTT assay; The morphologic changes of gastric cancer cells were observed and the mitosis index was assessed by inverted microscope; The ultratructure of gastric cancer cells were observed with the transmission electron microscope (TEM) ; TUNEL method was used to detect the apoptosis of gastric cancer cells. RESULTS: The inhibitive effect of Seh B plus 5-FU was significantly stronger than that of Sch B alone, in a dose-dependent manner; Cells were treated with medicine for 48 h, the mitosis index in the combina tion treated group ( 1.1±0.28)% was significantly decreased than Sch Bgroup (13.7±0.43)% and 5 FU group (5. 1±0.52)% (P〈0.05) ; Cells shrinkage, nuclear envelope disappears, nuclear chroma tin condensation and apoptotic body formation were observed by TEM, and all of which were most significant in the group of two drugs. TUNEL detection showed that the apoptosis rate in the combination treated group (73.9±5. 1)% was significant than Sch B group (16.4±2.4)% and 5-FU group (34. 6±3. 2)% (P〈0.05). CONCLUSION: The combination of Sch B and 5-FU show an effective activity in inhibiting gastric cancer cell proliferation and promoting cell apoptosis, and have a synergistic effect between them.
