摘要
目的探讨PDCD5、MDM2、Tip60和P53在新疆食管鳞癌患者的癌组织、癌旁组织和正常组织中的表达及相关性。方法 RT-PCR检测PDCD5、MDM2和Tip60 mRNA表达;免疫组织化学法检测PDCD5、MDM2、Tip60和P53蛋白的表达(18例哈萨克族、22例汉族)。结果癌组织中PDCD5、MDM2和Tip60 mRNA的阳性率为80.0%、65.0%和97.5%,MDM2 mRNA在癌组织中阳性率高于正常和癌旁组织(P<0.05);半定量RT-PCR结果显示癌组织PDCD5、MDM2、Tip60 mRNA低于正常组织(P<0.05)。癌组织PDCD5、MDM2和P53蛋白表达率低于正常组织(P<0.05);癌组织中PDCD5蛋白的阳性表达与分化程度有关(P<0.05);PDCD5与MDM2 mRNA、Tip60与P53蛋白在癌组织中的表达有相关性(P<0.05)。结论 PDCD5、MDM2、Tip60和P53四者之间可能存在共同的激活机制,构成了肿瘤细胞凋亡的多种途径。
Objective To study the relationship between expressions of PDCD5,MDM2,Tip60 and P53 in esophageal squamous cell cancers from Xinjiang cases.Methods RT-PCR was used to detect mRNA of PDCD5,MDM2,Tip60.The PDCD5,MDM2,Tip60 and P53 protein expressions were determined by using immunohistochemisry staining method.Results The positive incidence of PDCD5,MDM2 and Tip60 mRNA expressions in 40 cases of esophageal cancer tissues were 80.0%,65.0% and 97.5% respectively.The expression rate of MDM2 mRNA in esophageal cancer was significantly higher than that in normal esophagus mucosa and para-cancer tissue(P0.05).Semi-quantitative analysis showed expression of PDCD5,MDM2 and Tip60 mRNA were of statistical difference in esophageal cancer and normal esophagus mucosa(P0.05).Expression rate of PDCD5,MDM2 and P53 proteins were lower in esophageal cancer than that in normal esophagus mucosa(P0.05).The positive expression rate of PDCD5 protein in esophageal cancer was significantly correlated with tumor differentiation(P0.05).Theexpression of Tip60 and P53 protein,PDCD5 and MDM2 mRNA were significantly correlated(P0.05).Conclusions It is speculated that there may exist a common transcription mechanism among PDCD5、MDM2、Tip60 and P53,and constitute multiple pathways to induce cell apoptosis.
