摘要
目的:观察地骨皮醇提取液对2型糖尿病大鼠肾脏COX-2表达的影响,探讨其对糖尿病肾病大鼠肾脏保护作用的机制。方法:SD大鼠125只,随机抽取10只为正常对照组。以STZ加高脂饲料饲养3周复制2型糖尿病肥胖大鼠模型,随机分为模型组、罗格列酮组、地骨皮醇提低剂量组、地骨皮醇提高剂量组,每组10只。模型组和正常组给予生理盐水,各实验组分别给予相应药物灌胃,治疗7周后免疫组化法检测肾脏COX-2表达。结果:罗格列酮组和地骨皮醇提组COX-2表达与模型组比较明显改善,差异均有统计学意义(P<0.01),但与正常对照组之间仍有明显差异(P<0.01)。地骨皮醇提高剂量组与罗格列酮组比较差异无统计学意义(P>0.05)。结论:地骨皮醇提取液可以降低2型糖尿病肥胖大鼠肾脏COX-2的表达,其中高剂量组优于低剂量组,与罗格列酮组疗效相当。
To observe the effect of Digupi extract on renal expression of cox-2 in rats with type 2 diabetic mellitus(DM ), and to reveal the underlying mechanism. Methods: Sprague Dawley rats were given high fat diet for 3 weeks to establish type 2 DM obesity. Obese rats were randomly divided into model group, x'osiglitazone group, Digupi ethanol extract low dose group(low Digupi group), and Digupi ethanol extract high dose group(high Digupi group), 10 in each. Other 10 rats served as nm'mal control. The model and normal control groups were given saline and other groups were intragastrieally given respective drugs for 7 weeks. Then rats were sacrificed to detect the expression of cox-2 in the kidney by immunohistochemistry. Results: The expression of cox-2 was significantly reduced in the rosiglitazone group and Digupi groups as compared with that in the model group(P〈0.01 ), but it was still higher than that in the normal control group(P〈 0.01 ). No significant difference was noted between the rosiglitazone group and the high Digupi group(P〉0.05). Conclusion: Digupi ethanol extract can reduce the renal expression of cox-2 in type 2 diabetic rats. High dose of Digupi had a better effect, which was similar to the efficacy of rosiglitazone.
