被动免疫制剂抗Pg-IgY对大鼠实验性牙周炎的治疗 被引量:4

摘要 目的:观察抗Pg-IgY制剂对大鼠实验性牙周炎的疗效。方法:将15只大鼠分为抗Pg-IgY治疗组、米诺环素治疗组、PBS冲洗组(阴性对照)、模型组(空白对照)和健康对照组。除健康对照组外,其余各组采用丝线结扎加Pg菌接种建立牙周炎模型后,按分组给予相应药物治疗,每周治疗1次,共治疗4周,模型组不做任何处理。观察并记录治疗前后各组体质量、探诊深度、龈沟出血指数和松动度的变化,治疗4周后处死行HE染色观察病理变化。结果:建模和治疗期间各组大鼠一般情况良好,体质量稳定。抗Pg-IgY组和米诺环素组各项牙周临床指标均较治疗前明显改善(P<0.05);PBS组除松动度外,其他指标均与治疗前无显著差异(P>0.05)。各组间治疗后各项临床指标相比,抗Pg-IgY组与米诺环素组均无显著性差异(P>0.05),两组探诊深度和龈沟出血指数均较PBS组和模型组有显著性差异(P<0.05)。组织学观察结果显示抗Pg-IgY可控制局部炎症并存在一定的组织修复。结论:抗Pg-IgY能有效改善大鼠实验性牙周炎的临床症状,具有良好的治疗效果。 AIM: To evaluate the effects of egg yolk-derived immunoglobulin (IgY) against Porphyromonas gingivalis-induced experimental periodontitis in rats. METHODS: Fifteen male Sprague-Dawley rats were randomly divided into 5 groups, with 3 rats in each group. Periodontitis was induced by silk ligature technique, 100 g/L high sucrose drinking water and Porphyromonas gingivalis inoculation for 7 weeks in four groups, one group was left as periodontally healthy control. After periodontitts was estalished three groups were then treated with anti-P, g IgY (2mg/ mL), minocycline hydrochloride (2mg/mL) or PBS, once a week for 4 weeks, respectively. The other two groups served as controls. Mobility, was estalished, (MOB) of teeth, probing depth (PD) and sulcus bleeding index (SBI) of first molars were recorded at the end of the treatment. The rats were then sacrificed and histological assessment was carried out by HE staining. Data was analyzed by one-way analysis of variance (ANOVA) using SPSS 10.0. RESULTS : Periodontal parameters including MOB, PD and SBI were significantly improved in both anti-P, g lgY and minocycline hydrochloride groups at the end of treatment ( P 〈 0.05 ). However, no significant difference was found between anti-P, g IgY and minocycline hydrochloride. Histological examination also showed that anti-P, g IgY and mino cycline hydrochloride were effective in the treatment of chronic periodontitis. CONCLUSION: Anti-P. g IgY showed similar effects to minocycline hydrochloride in the treatment of chronic periodontitis.
