摘要
目的:探讨单核细胞趋化蛋白-1(MCP-1)在大鼠肾脏缺血再灌注中的表达及其药物干预。方法:雄性Wistar大鼠60只,随机分为6组。夹闭肾动脉建立大鼠肾缺血再灌注模型,在再灌注不同时点检测肾组织MCP-1的含量、血肌酐(Cr)、尿素氮(BUN)水平;光镜下观察肾脏的病理改变并进行肾小管评分;同时,观察药物洛伐他汀对上述指标变化的影响。结果:正常情况下,肾脏中MCP-1少量表达,缺血再灌注0h(单纯缺血)即可观察到肾组织中MCP-1表达升高,至4 h达到峰值,随后其水平逐渐下降。应用洛伐他汀干预后,肾组织MCP-1蛋白表达减少(P<0.01);血Cr、BUN比单纯缺血再灌注4 h组降低(均P<0.05);病理组织学肾小管评分降低(P<0.01)。结论:缺血再灌注后肾组织中MCP-1表达升高,提示MCP-1参与肾损伤的发生;洛伐他汀可能通过减少MCP-1的表达抑制炎症反应过程,减轻肾组织损伤。
Objective: To investigate the expression of monocyte chemoattractant protein-1 and drug intervening in ischemia / reperfusion rat kidney.Methods:Sixty male wistar rats were randomly divided into six groups.A model of renal I/R injury was made to clamp the kidney artery.Expression of MCP-1 was measured by ELISA after reperfusion.We calculated the concentration of serum Cr,BUN that can imply the renal function.The pathologic change of renal was observed with microscope.And to observe the effect of Lovastatin on these indexes about-mentioned.Results:Expression of MCP-1 in the kidney is a little normally.We could observe the expression of MCP-1 increased at 0 hour after I/R(just ischemia) and reached a peak value at 4 hour,then dropped off subsequently.After Lovastatin was administered,the expression of MCP-1 in renal reduced(P0.01);Serum Cr,BUN decreased respectively compared with I/R 4h group(P0.05);And pathologic Paller's grade diminished(P0.01).Conclusion:The expression of MCP-1 increased in the renal ischemia/ reperfusion.These imply that MCP-1 may relate with the injury of renal;Lovastatin could decrease the MCP-1,repress the inflammatory reaction and alleviate the renal function impaired in some extent.
