摘要
目的探讨肝脏部分缺血再灌注损伤对小鼠空间认知功能的行为学影响。方法昆明种小鼠90只,随机分为正常对照组(Con组)、假手术组(Sham组)、缺血40 min再灌注30 min组(IR组),每组30只。Con组小鼠正常饲养,不做任何处理。Sham组小鼠戊巴比妥麻醉后,腹部正中切口剖腹,分离肝十二指肠韧带,然后关腹,不做任何缺血处理。IR组小鼠于肝脏缺血40 min后再灌注30 min关腹,抗炎治疗3 d。IR组造模成功后,Con组随机分为C1、C2、C3组,每组10只;Sham组随机分为S1、S2、S3组,每组10只;IR组随机分为IR1、IR2、IR3组,每组10只。C1、S1、IR1组(即术后第4天组)小鼠于术后第4天行Morris水迷宫实验和避暗实验,C2、S2、IR2组(即术后第11天组)小鼠于术后第11天行Morris水迷宫实验和避暗实验,C3、S3、IR3组(即术后第18天组)小鼠于术后第18天行Morris水迷宫实验和避暗实验,Morris水迷宫实验持续7 d,避暗实验持续6 d。结果①Morris水迷宫实验定位航行试验:与S1组比较,C1组逃避潜伏期差异无统计学意义;IR1组于测试第2~7天逃避潜伏期高于S1组,差异有统计学意义(P<0.01)。与S2组比较,C2组逃避潜伏期差异无统计学意义;IR2组于测试第2、3天逃避潜伏期高于S2组,差异有统计学意义(P<0.05);与S3组比较,C3、IR3组逃避潜伏期差异无统计学意义。②Morris水迷宫实验空间搜索实验:与S1组比较,C1组穿环次数、平均速度、中心活动路程及中心活动时间差异无统计学意义;IR1组各指标均低于Sham组,差异有统计学意义(P<0.01)。术后第11、18天组各组的空间搜索实验比较,差异无统计学意义。③避暗实验:结果与Morris水迷宫趋势一致。结论肝脏部分缺血再灌注损伤可致小鼠术后短期认知功能障碍,认知功能障碍持续10 d左右,且这种认知功能障碍具有可逆性。
Objective To investigate the behavioral effects of partial hepatic ischemia reperfusion injury(HIRI) on postoperative cognitive function in mice.Methods Ninety mice were randomly divided into three groups: control group(C group,n=30);sham group(S group,n=30);IR groups(n=30): the mice underwent partial hepatic ischemia for 40 min followed by reperfusion for 30 min.Each group was further divided into 3 subgroups:C1,C2,C3,S1,S2,S3,IR1,IR2,IR3(n=10).Behavioral tests of C1,S1,IR1 were detected from the 4th day;and C2,S2,IR2 were detected from the 11th day;C3,S3,IR3 were detected from the 18th day.Morris water maze lasted for seven days,step through test lasted for six days.Results ① Compared with the latency period of Morris water maze of S1,there was no significantly statistical difference between C1 and S1,IR1 was significantly longer than that in S1 in days 2,3,4,5,6 and 7(P0.01);while in the 11th day groups,there was no significantly statistical difference between C2 and S2,IR2 was significantly longer than that in S2(P0.05) in days 2 and 3;and there was no significantly statistical difference among three groups of the 18th day groups.② Compared with the piercing numbers,average speed,central swimming distance and swimming time of morris water maze search of S1,there was no significantly statistical difference between C1 and S1,IR1 was obviously shorter than that in S1(P0.01),and there was no significantly statistical difference among three groups of the 11th and 18th day groups.③ The results of the step through test were consistent with the Morris water maze.Conclusion Partial HIRI leads to a short term postoperative cognitive dysfunction in mice,and which lastes about 10 days,and this cognitive dysfunction is reversible.
