亲和超滤技术快速发现中药中抗炎成分的方法学 被引量:4

摘要 目的建立一种生物亲和超滤方法解决抗炎活性物质快速筛选的问题。方法利用生物亲和超滤技术设计以COX2酶为靶酶,在中草药提取物中添加COX2酶抑制剂塞来昔布进行筛选,以格列吡嗪作为对照,结合HPLC-DAD和HPLC-MS技术进行快速分离和检测。结果成功筛选到目标化合物塞来昔布。结论亲和超滤与液质联用技术是快速、简便、高效、灵敏、成本相对较低的筛选模式。 Objective To set up a method of affinity ultrafiltration,to solve the fast screening of anti-inflammatory active compounds.Methods Utilizing affinity ultrafiltration design methods,and cyclooxygenase-2(COX-2)as target enzyme,we added celecoxib(a kind of COX-2 inhibitor)to the extractive from Chinese medicines for screening,compared with glipizide,to separate and detect active compounds combining together HPLC-DAD and HPLC-MS technology.Results This method could screen the target compound celecoxib successfully.Conclusion As a screening mode,this method has much advantages,such as quickness,convenience,high performance,sensitiveness,and correspondence lower cost.
