摘要
目的通过对大鼠肺组织中Toll样受体4(TLR-4)和转化生长因子β1(TGF-β1)表达影响的研究及对大鼠肺组织病理形态的观察,评价青蒿琥酯对哮喘大鼠的药理作用。方法清洁级SD雄性大鼠32只,随机分为对照组、哮喘组、地塞米松组和青蒿琥酯组,采用激发和雾化吸入的方法建立哮喘模型。观察各组大鼠肺组织病理改变;取相关组织分别进行外周血白细胞计数、肺泡灌洗液(BALF)白细胞计数;Westernblot法和RT-PCR法检测TLR-4和TGF-β1蛋白表达情况。结果与哮喘组相比较,青蒿琥酯组大鼠肺部组织的炎症细胞浸润较少,肺泡的组织结构较为完整;外周血和BALF白细胞计数结果表明,青蒿琥酯组各项指标与哮喘组比较均有所改善;在蛋白水平上,青蒿琥酯组TLR-4和TGF-β1蛋白表达均下降。结论青蒿琥酯逆转哮喘大鼠组织病理变化,降低TLR-4蛋白表达,并且可能通过TLR-4调整TGF-β1表达,最终改善哮喘大鼠的症状。
Objective To analyze the therapeutic effect of artesunate on the expression of TLR-4 and TGF-β in asthma rats. Methods The asthma model was established by ovalbumin (OVA) sensitizing-challenging SD rats. Thirty-two rats were randomly divided into four groups: the control group, the asthma model group, the dexamethasone treating group and artesunate treating group. The anti-asthmatic effects were assessed by histopathological examination and Western blot was applied to analyze the expression of TLR-4 and TGF-D in lung tissues. Results In asthma group, the membrana mucosa plica became multiple, the thickness of airway smooth muscle increased. A great quantity of EOS infiltrated to membrana mucosa, bronchi wall and pulmonary alveoli. Inflammatory reactions of dexamethasone treating group and artesunate treating groups were obviously extenuated. The expression levels of TLR-4 and TGF-β in artesunate treating group were reduced. Conclusion Artesunate can release asthma through decreasing the expression of TLR-4 and TGF-β1.
