摘要
目的探讨小儿麻藤定喘方有效成分群的水提醇沉工艺条件。方法以盐酸麻黄碱含量为指标,采用高效液相色谱法测定含量,运用正交试验法对水提及醇沉的工艺条件进行优化;采用乙酰胆碱和组胺混合液致豚鼠哮喘发作,观察哮喘潜伏期,通过药效验证最佳工艺。结果水提最佳工艺为第1次补足吸水量后加10倍量水浸泡2 h,其余每次加10倍量水提取3次,每次1.5 h;醇沉最佳工艺为醇沉浓度为60%,浓缩比例为1∶1.5,静置时间为24h;按此工艺条件提取的成分可明显延长哮喘发作潜伏期。结论优选的工艺稳定、可行。
Objective To optimize the process condition for the water extraction and alcohol precipitation of Xiaoermateng Dingchuanfang.Methods Orthogonal test was used to optimize the extraction and precipitation process.Using ephedrine hydrochloride as assessment index,quantification of extraction and precipitatio n was evaluated by HPLC and antiasthmatic effects of extract on asthma latency o f guinea pigs induced by the mixed solution of acetylcholine and histamine.Resul ts The optimum extraction process was as follows:10 times of water was added to soaked medicine material of Xiaoermateng Dingchuanfang and filtered after boili ng for 1.5 h.The extraction process was repeated for three times.The optimum alc ohol precipitation process was as follows:precipitation in 60%of aqueous alcoh ol for 24 h.The extracts in the optimum process conditions significantly prolong ed asthma latency induced by the mixed liquor of acetylcholine and histamine.Con clusion The optimum process is stable and feasible.
