己糖激酶法间接测定人血浆和尿样中的羟乙基淀粉浓度及药动学应用

Indirect Determination of Hydroxyethyl Starch in Human Plasma and Urine by Hexokinase Method and Its Application in Pharmacokinetic Study

摘要 目的建立一种快速、灵敏的方法间接测定人血浆和尿样中的羟乙基淀粉浓度。方法血浆和尿样经三氯乙酸沉淀蛋白,丙酮沉淀羟乙基淀粉后,以三氟乙酸水解羟乙基淀粉,采用己糖激酶法测定水解产物单糖,从而计算血浆和尿中羟乙基淀粉的浓度。结果测定血浆中羟乙基淀粉方法的线性范围为0.25~10.0 mg.mL-1,其涵盖实际剂量下血药浓度范围。方法确证各结果均符合相关标准。实验每天可以分析150个样品。结论本分析方法测试时间短,灵敏度高,血浆用量少,适用于人血浆和尿样中羟乙基淀粉的测定及其药动学研究。 OBJECTIVE To develop a rapid and sensitive analytical method for quantification of hydroxyethyl starch in hmnan plasma and urine. METHODS The hydroxyethyl starch was precipitated by actone after deproteinization of the plasma and urine sample with trichloroacetic acid. After the removal of the supernatant, the hydroxyethyl starch precipitate was dissolved and hydrolyzed into monomer with trifluoroacetic acid. After drying and dissolving in phosphate buffer solution, the glucose determination was performed using an enzymatic test based on hexokinase/glucose 6-phosphatese, and the concentration of hydroxyethyl starch was calculated according to the concentration of glucose. RESULTS The linear range of the calibration curve for hydroxyethyl starch was 0. 25 - 10. 0 mg·mL^-1 which covered the actual plasma concentration. The intra-day and inter-day relative standard deviations of all the check samples were less than 13.2% , and the relative error was in the range of -2. 0% to -0. 7%. The assay procedure has been validated in term of accuracy, precision, linearity, selectivity and robustness. 150 samples could be analysed per day. CONCLUSION The method is proved to be sensitive, rapid and suitable for the determination of hydroxyethyl starch in human plasma and urine as well as the investigation of hydroxyethyl starch pharmacokinetics.
