摘要
目的探讨前列地尔对局灶性脑缺血大鼠大脑皮层磷酸化c-Jun氨基末端激酶(p-JNK)和磷酸化c-Jun(p-c-Jun)表达的影响。方法将30只健康大鼠随机分为假手术组(A组)、缺血对照组(B组)、前列地尔治疗组(C组)、阳性药物对照组(D组),其中C组又分为1.25、2.50、5.00μg/kg三个亚组,每组5只。采用大脑中动脉闭塞法制作局灶性脑缺血模型,大鼠清醒后行神经功能缺损评分;应用免疫组化法检测其大脑皮层缺血6 h后的p-JNK、p-c-Jun表达。结果 C、D组神经功能缺损评分明显低于B组(P均<0.05)。A组大脑皮层偶见p-JNK、p-c-Jun阳性细胞,B组可见大量阳性细胞;与B组比较,C组各亚组p-JNK、p-c-Jun阳性细胞数明显降低(P均<0.05)。结论前列地尔预处理对大鼠局灶性脑缺血有保护作用,其机制可能与抑制脑组织p-JNK、p-c-Jun表达有关。
objective To study the effects of prostaglandin E1 on the expressions of phospho-c-Jun NH-terminal kinase(p-JNK),phosphorylated c-Jun(p-c-Jun) in brain tissue after focal cerebral ischemia in adult rats.Methods A total of 30 healthy rats were divided into sham group(group A),ischemia control group(group B),prostaglandin E1 treatment group(group C),positive medicine group(group D).C group was divided into 1.25,2.50,5.00 μg/kg subgroup.Cerebral ischemia models were made by middle cerebral artery occluding in rats.After focal cerebral ischemia for 6 hours,the neurological deficit score was evaluated and immunohistochemistry technique was used to observe the expressions of phosphorylated JNK,phosphorylated c-Jun.Results Compared with group B,the neurological deficit scores were lower in group C,D,p-JNK,p-c-Jun positive cells were decreased in all subgroups.Fewer p-JNK or p-c-Jun positive cells were existed in cerebral cortex in group A,and lots of them were existed in group B(all P0.05).Conclusions Prostaglandin E1 can protect the brain cells from damage after focal cerebral ischemia injury by adjusting the expressions of phosphorylated JNK and phosphorylated c-Jun.
