摘要
建立了水解胶原蛋白口服液中肽分子量分布的基质辅助激光解吸/电离飞行时间质谱(MALDI-TOF-MS)检测方法。实验探讨了基质的选择、基质与样品的配比及检测方式对测定的影响,结果表明,采用合适的基质与样品的配比,在正离子反射模式下,以2,5-二羟基苯甲酸(DHB)为基质,可得到水解胶原蛋白口服液中肽分子量分布的质谱图,该水解胶原蛋白口服液中肽的分子量主要集中在0.6~1.5ku。该方法简单快速,有较高的准确性,且提高了检测效率,可实现批量检测。
The molecular weight distribution of peptides in hydrolyzed oral collagen was determined by matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF-MS).The effects of the matrix,the appropriate proportion of sample/matrix and the method of detecting were disscussed.The conclusion is that the molecular weight distribution of peptides in hydrolyzed oral collagen was obtained in the appropriate proportion of sample/matrix the reflection positive mode and using DHB as the matrix.The molecular weight distribution of peptides in hydrolyzed oral collagen is between 0.6 and 1.5 ku.The method is simple and convenient,with high accuracy and high detection efficiency.This method can achieve batch testing.
