摘要
目的:建立15种常见精神类药物在人体使用过程中的监测条件,即蛋白沉淀-高效液相色谱法联合分析法的可行性。方法:0.5 mL血浆样品加入1.5 mL乙腈,旋涡混合后,离心,上清液过滤,滤液直接采用HPLC测定。色谱条件:C18柱,采用梯度洗脱方式,流动相组成为:A.乙腈;B.0.1%磷酸水溶液。梯度洗脱程序:0.0~30.0 min(5%A~45%A,95%B~55%B),30.0~35.0 min(45%A~80%A,55%B~20%B),流速1.0 mL.min-1,检测波长210 nm,进样量20μL,柱温30℃。结果:15种药物平均回收率均大于80%,RSD 0.44%~8.07%。结论:该法乙腈沉淀蛋白的条件简单,具有快速、回收率高且稳定,能够作为血浆中药物检测分析的通用前处理方法;梯度洗脱可以作为血浆样品的首选常规色谱分析方法。
OBJECTIVE To establish a protein precipitation-HPLC method for determination of 15 common-used drugs in plasma. METHODS 0. 5 mL plasma was added with 1.5 mL acetonitrile, vortex mixed and centrifuged, the supernatant was filtrated and then injected into the HPLC system. The HPLC conditions are detailed as following: with C18 column, mobile phase of acetonitrile-0. 1% phosphate water solution by gradient elution, 0. 0 - 30. 0 min(5 % A- 45 % A, 95%B- 55 %B), 30. 0 + 35.0 min(45%A- 80%A,55%B- 20%B), the flow rate was 1.0 mL·min^-1. The detection wavelength was set at 210 nm, and the injection volume was 20μL, column temperature was 30℃. RESULTS The average absolute recoveries of 15 drugs tested exceeded 80% with RSD ranging from 0. 44% to 8. 07%. CONCLUSION The results showed that protein precipitation by acetonitrile is suitable for plasma drug analysis.
