摘要
目的采用HPLC法测定麻花艽地上和地下部位中马钱苷酸、獐牙菜苦苷、龙胆苦苷和獐牙菜苷的含量。方法采用Eclipse XOB-C18色谱柱(150 mm×4.6 mm,5μm);流动相为甲醇-水(21∶79);流速1 mL.min-1;检测波长254 nm;柱温25℃。结果马钱苷酸、獐牙菜苦苷、龙胆苦苷和獐牙菜苷的线性范围分别为0.08~6.31μg(r=0.9997)、0.069~4.12μg(r=0.9999)、0.06~6.23μg(r=0.9998)、1.32~7.76(r=0.9999);平均回收率分别为99.7%(RSD=2.5%)、101.2%(RSD=1.9%)、102.4%(RSD=1.6%)、100.5%(RSD=2.2%)。结论所建方法测定快速、结果准确;麻花艽地上部位可作为主要的药源。
OBJECTIVE To establish an HPLC method for the determination of loganic acid,swertiamaroside glycosides,gentiopicroside and swertiamar in ground and underground parts of Gentiana straminea.METHODS Eclipse XOB-C18(150 mm×4.6 mm,5 μm)column served as the solid phase,the mobile phase was methanol-water(21:79).The flow rate was 1 mL·min-1,the detection wavelength was 254 nm,temperature was 25 ℃.RESULTS The linear range of loganic acid,swertiamaroside glycosides,gentiopicroside and swertiamarin were 0.08-6.31 μg(r=0.9997),0.069-4.12 μg(r=0.9999),0.06-6.23 μg(r=0.9998)and 1.32-7.76(r=0.9999),respectively.The average recovery were 99.7%(RSD=2.5%),101.2%(RSD=1.9%),102.4%(RSD=1.6%)and 100.5%(RSD=2.2%),respectively.CONCLUSION The method is rapid,accurate and reliable.Gentiana straminea ground parts can be used as important sources of medicine.
