摘要
目的:建立怀牛膝药材中5-羟甲基糠醛和蜕皮甾酮的高效液相色谱测定方法。方法:SHIMADZU shim-pack VP-ODS(4.6 mm×250 mm,5μm)柱,流动相:乙腈-水梯度洗脱,流速:1.0 mL.min-1,柱温:30°C,检测波长:245nm和279 nm。结果:可在2个波长下同时测定5-羟甲基糠醛和蜕皮甾酮的含量,5-羟甲基糠醛的线性范围是0.390~100μg.mL-1,蜕皮甾酮的线性范围是3.125~100μg.mL-1。平均回收率分别为100.4%和99.0%。结论:该方法灵敏准确,为怀牛膝药材的质量控制提供了新的含量测定方法。
Objective:To establish an HPLC method for the simultaneous determination of the contents of 5-hydroxymethylfurfural and ecdysterone in Achyranthes bientatae B1.Methods:A SHIMADZU shim-pack VP-ODS(4.6 mm×250 mm,5 μm) column was adopted,with a mixture solution of Acetonitrile-water as mobile phase using a gradient elution at flow rate of 1.0 mL·min-1,the column temperature was 30 ℃,and the detection wavelength was 279 nm and 245 nm.Results:The contents of two compounds were simultaneously detected at two wavelengths.The calibration curves showed good linearity in the range of 0.39-100 μg·mL-1(5-hydroxymethylfurfural) and 3.125-100 μg·mL-1(ecdysterone),and the average recoveries of the method were 100.4% and 98.99%.Conclusion:The method is sensitive and accurate,and can be used for the quality control of Achyranthes bientata B1.
