摘要
目的建立甲状腺球蛋白抗体(TgAb)时间分辨荧光免疫分析(TRFIA)间接检测法。方法用Tg抗原包板,用铕(Eu3+)标记兔抗人IgG做标记物,间接法TRFIA检测人血清中的TgAb。结果 TgAb-TRFIA的灵敏度为1.0 IU/ml;批内变异系数(CV)为3.1%~3.6%,批间CV为3.3%~3.6%;平均回收率为101.6%;热稳定性好;与电化学发光分析技术(ECLIA)比对,相关系数达0.8945;与临床结果高度相关。结论本法建立的TgAb-TRFIA是一个高灵敏和可靠的检测,有助于甲状腺疾病的临床诊断。
An indirect time-resolved fluoroimmunoassay(TRFIA) for thyrogloblin antibody(TgAb) was established and its clinical application was evaluated.The Tg antigen was coated on the 96 well plates.A rabbit anti-human IgG-Eu3+ was used to enable detection.Serum TgAb level was detected with the kit by TRFIA.Assay sensitivity was 1.0 IU/ml.Intrassay coefficient of variation was 3.1%~3.6%.Interassay coefficient of variation was 3.3%~3.6%.Average recovery was 101.6%.The kit was stable.Correlation coefficient with TgAb ECLIA was 0.8945.The results were highly relevant to clinical,which indicated that the indirect TRFIA for TgAb detection in serum is a sensitive and reliable assay,and would be helpful to clinical diagnose for thyroid diseases.
