外源性TGF-β1诱导人乳腺癌MCF-7细胞的乙醛脱氢酶1 mRNA及蛋白表达 被引量:3

摘要 目的本研究旨在探讨外源性TGF-β1诱导人乳腺癌MCF-7细胞对乙醛脱氢酶1(aldehyde dehydrogenase1,ALDH1)mRNA及蛋白表达的影响。方法用不同浓度(1、5ng/mL)的TGF-β1诱导体外培养的MCF-7细胞48h后,RT-PCR检测ALDH1mRNA的表达变化,Western blot检测ALDH1蛋白的表达变化。结果 TGF-β1(1、5ng/mL)诱导的MCF-7细胞的ALDH1mRNA及蛋白表达均高于未诱导的MCF-7亲本细胞,差异有统计学意义(P均<0.001);高浓度(5ng/mL)TGF-β1诱导的MCF-7细胞的ALDH1mRNA及蛋白表达均高于低浓度(1ng/mL)TGF-β1诱导的MCF-7细胞,差异有统计学意义(P均<0.001)。结论外源性TGF-β1可诱导人乳腺癌MCF-7细胞上调ALDH1mRNA及蛋白的表达,可能诱导ALDH1+乳腺癌肿瘤干细胞(cancer stemcell,CSC)的转化,并有一定的浓度依赖性。 Objective To investigate the effects of exogenous TGF-β1 on the regulation of aldehyde dehydrogenase 1(ALDH1) expression in human breast cancer MCF-7 cells.Methods MCF-7 cells were induced with TGF-β1 at different concentrations(1,5 ng/mL) for 48 hours.ALDH1 expression in these cells was then analyzed at mRNA and protein levels by RT-PCR and Western blot assay,respectively.Results ALDH1 mRNA and protein expressions were up-regulated in TGF-β induced(1,5 ng/mL) MCF-7 cells compared with normal MCF-7 cells,respectively(All P0.001).They were also up-regulated in MCF-7 cells induced with TGF-β1 at higher concentration(5 ng/mL) compared with MCF-7 cells induced with TGF-β1 at lower concentration(1 ng/mL),respectively(All P0.001).Conclusions Exogenous TGF-β1 up-regulats expressions of ALDH1 mRNA and protein in human breast cancer MCF-7 cells in the manner of concentration-dependence,and TGF-β1 may mediates the convention of cancer stem cells(CSC) to breast cancer cells.
