摘要
采用随机扩增多态性DNA技术对31种石斛属植物的遗传多样性进行分析.从100条RAPD引物中共筛选出18条引物,18条引物共扩增出1 802条带,其中多态性位点有261个,多态性比为99.2%.UPGMA聚类结果显示,基因型之间的相似系数分布于0.6420.838之间,31种石斛属植物样品分为7类.聚类分析显示供试样品完全被区分开,实验结果与传统形态分类相似,但又有一定区别.丰富稳定的扩增多态性证明RAPD标记技术可以用于石斛属植物的物种鉴定和遗传多样性研究.
The experiment analyzed the genetic diversity among 31 species of Dendrobium with Random Amplified Polymorphic DNA(RAPD).In total,1802 bands were amplified by 18 RAPD primers,resulting from 261 RAPD loci with 99.2% polymorphism at genus level.Unweighted pair group mean analysis(UPGMA) show that the genetic similarity ranged from 0.642 to 0.838,and the materials used in the experiment are grouped into seven clusters.The cluster analysis show that the materials are completely separated,the result of the experiment is basically similar to the traditional morphological classification,but there are some differences.The high polymorphism and reliable amplification across species demonstrate that RAPD can be used to identify the species of Dendrobium and genetic diversity.
