银杏酮酯对过氧化氢诱导大鼠乳鼠心肌细胞凋亡的抑制作用 被引量:1

Protective Effects of Ginkgo Biloba Extract 50 Against H_2O_2-induced Apoptosis in Neonatal Rat Cardiomyocytes

摘要 目的探讨银杏酮酯(Ginkgo biloba extract 50,GBE 50)对过氧化氢诱导的大鼠乳鼠心肌细胞凋亡的抑制作用。方法利用体外培养模型,以过氧化氢0.2 mmol.L-1诱导心肌细胞损伤。乳大鼠心肌细胞分为正常对照组、过氧化氢组、过氧化氢+银杏酮酯0.2 mg.mL-1组。用MTT法检测心肌细胞存活率;Western Blot方法检测Bcl-2、Bax和Caspase-3的蛋白表达水平。结果与正常对照组相比,过氧化氢组使心肌细胞的存活率显著降低(P<0.01),Bcl-2的蛋白表达水平明显降低(P<0.01),而Bax和Caspase-3蛋白表达水平显著增加(P<0.01),Bcl-2/Bax的比率降低(P<0.01);与过氧化氢组相比,过氧化氢+银杏酮酯组中,心肌细胞的存活率升高显著(P<0.01),银杏酮酯使Bcl-2的蛋白表达水平明显升高,Bax和Caspase-3的蛋白表达水平明显降低(P<0.01),Bcl-2/Bax的比率升高(P<0.01)。结论银杏酮酯对过氧化氢诱导的乳大鼠心肌细胞凋亡具有抑制作用。 Objective To investigate the protective effects of Ginkgo Biloba Extract 50 ( GBE 50) against H2O2 - induced apoptosis in neonatal rat cardiomyocytes. Methods Myocardial cells of neonatal rats were cultured in vitro, 0. 2 mmol · L^- 1 H2O2 was used to induce cardiomyocyte apoptosis. Cultured neonatal rat cardiomyocytes were divided into three groups, i. e. control group, H2O2 group, and H2O2 plus 0. 2 mg·mL^-1 GBE 50 group. The eardiomyocyte viability was analyzed by MTT assay. The expression of Bcl - 2, Bax and Caspase - 3 were determined by Western blot. Result Compared with control group, H2O2 significantly decreased the viability of cardiomyocytes (P 〈 0.01 ), expression levels of Bel - 2 was down regulated ( P 〈 0. 01 ) , whereas the expression level of Bax and cleaved Caspase - 3 were significantly unregulated, and Be1 - 2/Bax ratio was decreased (P 〈 O. 01 ). Compared with H2O2 group, eardiomyoeyte viability was significantly increased in H2O2 + GBE 50 group (P 〈 0. 01 ) , GBE 50 treatment unregulated the expression level of Bel - 2, down regulated expression levels of Bax and cleaved Caspase - 3 ( P 〈 0. 01 ) , increased Bel - 2/Bax ratio ( P 〈 0. 01 ). Conclusion Taken together, these results suggest that GBE 50 could protect cultured neonatal rat cardiomyocytes against H2O2 -induced apoptosis.
