摘要
目的探讨低氧诱导因子-1α(HIF-1α)在类风湿性关节炎(RA)组织中的表达及其相关性作用。方法通过组织形态学和免疫学等手段检测,观察Ⅱ型胶原诱导性关节炎大鼠模型的关节组织中HIF-1α蛋白的表达情况。结果Ⅱ型胶原诱导性关节炎大鼠的关节滑膜层和滑膜下层均表达HIF-1α,第21天阳性表达量最高,随病程进展,表达逐渐下降,与滑膜病理学评分、滑膜增生评分及血管生成评分呈显著正相关。结论 HIF-1α在RA组织中的表达,与炎症严重程度呈正相关,提示HIF-1α对RA的发生发展密切相关,为RA的临床治疗提供了新的思路。
【Objective】 To study the expression of hypoxia-inducible factor-1α(HIF-1α) in rheumatoid arthritis(RA) tissue and its correlation.【Methods】 Through the means of morphological and immunological detection,joint tissue HIF-1α protein expression of type Ⅱ collagen-induced arthritis rat model was observed.【Results】 The synovial membrane and the lower synovial of rat joints in type Ⅱ collagen-induced rheumatoid arthritis both were expressed in HIF-1α.The positive expression of HIF-1α reached the highest volume in the first 21 days.Then with the disease progression,the expression gradually decreased,which was significantly positive related with the synovial pathology score,synovial hyperplasia and angiogenesis rating score.【Conclusion】 The expression of HIF-1α in RA tissue is positively related with inflammation severity.It indicates that HIF-1α is closely related to the occurrence and development of RA.The study will provide new ideas for the clinical treatment of RA.
