外用绞股蓝对自然衰老小鼠皮肤组织中CAT活性、MDA含量影响的实验研究 被引量:5

摘要 [目的]观察绞股蓝提取液对自然衰老小鼠皮肤组织中过氧化氢酶(CAT)活性及丙二醛(MDA)含量的影响,并研究不同的给药途径对自然衰老小鼠皮肤组织的改善作用。[方法]昆明种3月龄雌性小鼠10只,作为青年对照组(A);昆明种12月龄雌性小鼠40只,随机分为4组,每组10只:老年外用空白组(B)、老年外用阳性对照组(C)、老年外用绞股蓝组(D)、老年(外用+内服)绞股蓝组(E)。A、B组外涂赋型剂0.5g/t,C组外涂维生素E20mg/t,D组外涂绞股蓝霜剂0.5g/t,E组外涂绞股蓝霜剂(剂量同D组),同时灌胃绞股蓝提取液0.8mL/t。30天后以生化分析方法测定皮肤组织中CAT活性、MDA含量,并观察皮肤组织结构改变。[结果]与A组比较,B、C、D、E组小鼠皮肤组织CAT活性均明显降低(P<0.01),MDA含量明显增加(P﹤0.01);与B组比较,C、D、E组CAT活性提高(P﹤0.05),MDA含量降低(P﹤0.01);与D组相比,E组CAT活性明显提高(P﹤0.05),MDA含量明显降低(P﹤0.05)。[结论]绞股蓝提取液具有拮抗小鼠皮肤自然衰老的作用;外用配以灌胃绞股蓝提取液该种给药方式能更好的起到延缓自然衰老对小鼠皮肤的损害。 [Objective]To observe the gynostemma extract natural aging mice skin tissue catalase(CAT) activity and malondialdehyde(MDA) content,and to study different routes of administration of the natural aging mice skin improvement.[Method]3 month old female Kunming mice,10 rats as young control group(A);Kunming 40 female mice aged 12 months were randomly divided into 4 groups of 10 each: external control group aged(B),aged external positive control group(C),old external Gynostemma group(D),elderly(topical + oral) Gynostemma group(E).A,B group treated with excipient 0.5g / t,C group treated with vitamin E20mg / t,D group treated with Jiaogulan cream 0.5g / t,E group treated with Jiaogulan cream(the same dose of D),while ig Gynostemma extract 0.8mL / t.After 30 days determined by biochemical analysis of CAT activity in skin tissues,MDA,and to observe changes in the organizational structure of skin.[Results]A group,B,C,D,E groups of mice skin tissue CAT activity was significantly decreased(P0.01),MDA was significantly increased(P0.01);with the B group,C,D,CAT activity in group E increased(P0.05),MDA content decreased(P 0.01);with the D group,E group significantly increased CAT activity(P0.05),MDA content was decreased(P0.05).[Conclusion] Gynostemma extract skin of mice with the antagonistic effect of natural aging;external accompanied by intragastric administration of Gynostemma extract this kind of approach to better delaying the natural aging on skin damage.
