摘要
[目的]快速、有效、大批量地提取藻红蛋白。[方法]以坛紫菜为原料,采用溶胀法、化学试剂法和搅切法3种粗提技术,以藻红蛋白的提取得率、纯度和提取时间为参数对3种方法进行比较。[结果]溶胀法采用0.01mol/LTris-HCl(pH8.0)浸提9h,藻红蛋白得率为9.71mg/g,纯度(A565/A280)为0.45;化学试剂法在0.100mol/LSDS溶液作用4h时,藻红蛋白得率为24.35mg/g,纯度(A565/A280)为0.22左右;搅切法采用搅切速度18000r/min作用6min,藻红蛋白得率达到23.11mg/g,纯度(A565/A280)为0.30。[结论]综合比较,搅切法提取藻红蛋白效率高,藻红蛋白得率和纯度较好,处理量较大,是一种十分有效的粗提方法。
[Objective]The method of rapid,effective and a large amount of phycoerythrin extracted from Porphyra haitanensis was experimented.[Method]The indicator of the yield and purity of phycoerythrin from the Porphyra haitanensis and the extracting time in three kinds of the extraction techniques:swelling,chemical permeabilization and cutting while stirring were compared.[Result]The result showed that the yield of phycoerythrin from the Porphyra haitanensis was 9.71 mg/g,which purity was 0.45,with swelling technique under the condition of the concentration of Tris-HCl:0.01 mol/L,the pH value:8.0 and the extracting time:9 hours;the yield of phycoerythrin was 24.35 mg/g,which purity was about 0.22,with chemical permeabilization under the condition of the concentration of SDS:0.100 mol/L and the action time:4 hours and the yield of phycoerythrin was 23.11 mg/g,which purity was 0.30,with the cutting while stirring method under the condition of the stirring rate:18 000 r/min and the working time:6 minutes.[Conclusion]Based on the integrated comparison,the the cutting while stirring method was best in the extracting efficiency of phycoerythrin including its yield and purity and also a large amount of raw material could be treated at same time,which was an effective method.
