Quantitation of clozapine in human plasma by LC-MS/MS

摘要 建立了人血浆中氯氮平的固相萃取及LC-MS/MS定量检测方法。血浆中的氯氮平用固相萃取柱(Bond Elut Certify)萃取,采用Waters AtlantisTM d C18色谱柱,电喷雾离子源,正离子检测,多反应监测方式进行定量分析,SKF525a作为内标。对氯氮平的定量下限为100 ng/mL,氯氮平质量浓度在100~5000 ng/mL范围内线性良好(r≥0.9990),日内、日间精密度RSD≤15%(n=5)。方法适用于法庭与临床毒物分析。 To develop a method for quantitative determination of clozapine in human plasma by solid phase extr-action(SPE) and liquid chromatography-tandem mass spectrometry(LC-MS/MS).The plasma clozapine was extr acted using a solid phase extraction(SPE) and separated by Waters AtlantisTM d C18 column(150mm×3.9mm,5μ m).Electrospray ionization source was applied,positive ion mode was used to detect and multiple reaction monitorring mode was used to quantify clozapine.SKF525a was the internal standard.The limit of quantitation(LL OQ) for clozapine is 100ng/mL.This method allowed the reproducible and accurate quantification with the conc-entration range of 100-5000ng/mL.Intra-day RSD and inter-day RSD were less than 15%(n=5).This method seems to be quite specific,sensitive and accurate,and can be used to detect clozapine in forensic and clinical toxicological analysis.
