摘要
目的:探讨党参、黄芪、丹参等益气活血药对心梗后心功能不全大鼠心肌细胞RYR2,SERCA2a,PLB mRNA表达的干预作用。方法:SD大鼠42只,冠状动脉前降支结扎术后随机分成3组:假手术组(对照组)、模型组、中药组。模型组和对照组给予生理盐水灌服。中药组给予益气活血中药浸膏灌胃,治疗4周和8周后分别杀死一半大鼠,取出心脏PCR检测RYR2、SERCA2a及PLB mRNA。结果:各组心衰大鼠RYR2、SERCA2a及PLB mRNA的表达较对照组下降,但中药干预4周后中药组RYR2、SERCA2a及PLB mRNA表达较模型组都有明显增高(P<0.05)。中药干预8周后,RYR2、SERCA2a mRNA仍然高于模型组(P<0.05)。结论:党参、黄芪等中药能抑制心梗后心衰大鼠RYR2、SERCA2a及PLB mRNA下降的作用,但缺乏特异性,可能与益气活血药能促进心肌细胞的能量代谢,防止心衰后心肌细胞的凋亡,心室重构有关。
Objective: To explore the effect of invigorating vital energy (YIQI) and promoting -circulation (HouXue) herbs on expression of RYR2, SERCA2a, PLB mRNA in rat myocardial cells. The Yiqi-Huoxue formula includes Dangshen, Huangqi, Danshen and so on. Methods: 42 myocardial infracted (MI) rats were divided into 3 groups: normal group, model group, herbal group. The model group, and normal group were given normal saline; herbal group were given Yiqi and Huoxue herbs. RYR2, SERCA2a, PLB mRNA expression of all group rats myocardial cells were detected after four and eight week's medication. Results: The expression of RYR2, SERCA2a and PLB mRNA of all the groups of heart failure rat myocardial cells was decreasing after 4 week of medication, but that of herb group was higher than model group (P〈0.05). After 8 weeks of herb treatment, RYR2, SERCA2a mRNA was still higher than that of the model group (P〈0.05). Conclusion: The research results suggested that Yiqi- Huoxue herbs can inhibit the decrease of RYR2, SERCA2a and PLB mRNA in rat myocardial cells tbut not specificity. It may be related to the mechanism of Yiqi-Huoxue formula in promote the energy metabolism of myocardial cells and prevent apoptosis and ventricular remodeling.
