摘要
【目的】探讨缺氧诱导因子-1α(HIF-1α)单克隆抗体对子宫内膜异位症(EMs)模型的严重联合免疫缺陷病(SClD)小鼠病灶体积及组织微血管密度(MVD)影响。【方法】将子宫内膜组织接种于SClD小鼠腹部皮下,建立EMSSCID小鼠模型,随机分三组:对照组10例、生理盐水组10例和治疗组10例。对照组常规饲养,生理盐水组和治疗组分别在病灶内注射生理盐水和HIF—-α单克隆抗体,1周后重复注射1次,第2次治疗后1周处死三组SCID小鼠,测量EMS病灶的体积及免疫组化法测定病灶MVD的表达。【结果】治疗组的EMS病灶体积和MVD均显著小于生理盐水组及空白对照组(P〈0.05)。【结论】HIF-1α单克隆抗体治疗使SCID小鼠EMS病灶体积缩小,MVD明显减少,为临床治疗提供理论依据。
[Objective]To explore the effect of HIF-1α monoclonal antibody on lesion volume and microvessel density (MVD) of endometriosis(EMS) in serious combined immunodeficiency disease(SCID) mice. [Methods]The endometrium was transplanted into the skin in SCID mice to establish the SCID model of EMS. The mice were randomly assigned to control group( n = 10), normal saline group( n = 10) and treatment group( n =10). The control group was given routine feeding. Normal saline group and treatment group received intralesional injection with normal saline and HIF-1α monoclonal antibody, respectively. The injection was repeated one week later. All SCID mice were killed one week after the second injection. Lesion volume of EMS was measured. The expression of MVD was detected by immunohistochemical method. [Results] Lesion volume of EMS and MVD in treatment group were lower than those in normal saline group and control group( P 〈0.05). [Conclusion] HIF-1α monoclonal antibody therapy can reduce the lesion volume of EMS in SCID model and obviously decrease MVD, which provides the theoretical basis for clinical treatment.
