摘要
目的:观察雄黄体外抗人肝癌BEL-7402细胞作用和作用机制。方法:采用MTS法和细胞克隆实验法观察雄黄对BEL-7402细胞增殖的影响,应用PI/Hoechest33258双染法和流式细胞仪检测细胞凋亡,荧光分光光度计检测细胞内钙和线粒体膜电位。结果:与阴性对照组比,雄黄能剂量依赖性地抑制BEL-7402细胞的生长和克隆形成。雄黄作用后,BEL-7402细胞出现了凋亡形态学改变如染色质浓缩等;流式细胞仪检测显示,雄黄能剂量依赖性地诱导细胞凋亡,其中,225μg/ml雄黄引起BEL-7402细胞凋亡的百分率为(28.6±4.6)%。与阴性对照组比,雄黄作用后,BEL-7402细胞的细胞内钙升高和线粒体膜电位降低。结论:雄黄具有一定的抗肿瘤作用,其作用机制与升高细胞内钙、降低细胞线粒体膜电位并诱导细胞凋亡有关。
Objective:To investigate anticancer effect of realgar on human hepatocarcinoma cell line BEL-7402 in vitro.Methods:MTS assay and colony formation assay were used to elucidate the antiproliferative effect of realgar on BEL-7402 cells.PI/Hoechest33258 double dying and flow cytometer were carried out for cell apoptosis detection.Intercellular calcium concentration and mitochondrial membrane potential were detected using spectrofluorophotometer.Results:Realgar inhibited cell growth and colony formation of BEL-7402 cells in a dose-dependent manner when comparing with control group.Morphological changes chromatin condense of cell apoptosis were observed in realgar treated BEL-7402 cells,and a dose-dependent apoptosis was shown by flow cytometer analysis.Apoptotic percentage of BEL-7402 cells treated with 225 μg/ml realgar was(28.6±4.6)% and higher than that in the control.Furthermore,intercellular calcium was increased and mitochondrial membrane potential was decreased inBEL-7402 cells treated by realgar compared with control.Conclusion:Realgar has anticancer effect,which might be associated with increasing intercellular calcium,decreasing mitochondrial membrane potential and inducing apoptosis.
