摘要
以金线莲茎段、叶片、茎片、不定芽为试材,分别在添加6-BA、ZT、NAA、KT 5个不同处理的MS及1/2MS培养基上培养,诱导愈伤组织。结果表明,以茎段、茎片、不定芽为外植体,在MS+6-BA 2.0 mg/L+NAA0.5 mg/L、MS+NAA 2.0 mg/L+KT 0.1 mg/L和MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+ZT 0.2 mg/L培养基中培养,均能成功诱导愈伤组织。不定芽为诱导愈伤组织最佳外植体,最佳培养基为MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+KT 0.2 mg/L。
With the stems,leaves,segments,buds of Anoectochilus roxburghiias as explants,the induction of callus was studied by culturing at 5 different culture mediums with 6-BA,ZT,NAA,KT respectively.The results showed that explants including stems,segments and buds could successfully induce callus occurrence at following culture mediums,MS + 6-BA 2.0 mg/L + NAA 0.5 mg/L,MS + NAA 2.0 mg/L + KT 0.1 mg/L and MS + 6-BA 2.0 mg/L + NAA 0.5 mg/L + ZT 0.2 mg/L.Certainly,6-BA,NAA,KT were the important factors on production of callus.Among the three explants,the buds were the best explants for the callus induction.And the results of orthogonal design showed that for the bud of Anoectochilus roxburghii to form the callus,the best medium was MS + 6-BA 2.0 mg/L + NAA 0.5 mg/L + KT 0.2 mg/L.
