LPS诱导大脑皮质胶质细胞自噬基因Beclin-1蛋白表达的研究 被引量:1

摘要 目的:探讨脂多糖(lipppolys accride,LPS)刺激对大脑皮质胶质细胞自噬相关基因Beclin-1蛋白表达的影响。方法:体外分离新生小鼠大脑皮质混合胶质细胞,采用1 μg/ml LPS刺激24h后收集细胞与培养上清。Western blotting检测Beclin-1蛋白表达水平;CCK-8试剂盒检测细胞增殖率。结果:LPS刺激后,大脑皮质混合胶质细胞Beclin-1蛋白表达水平增高,与对照组比较差异具有统计学意义(P<0.05);细胞增殖率下降,与对照组比较差异亦具有统计学意义(P<0.05)。结论:LPS能激活神经胶质细胞自噬相关基因Beclin-1,这种变化参与调节了胶质细胞的存活情况。 Objective:To study the influence of LPS stimulation on expression of Beclin-1 protein.Methods:Cerebral mixed glia were isolated and cultured in vitro.With 1 μg/ml of LPS-stimulation,mixed glia and cultured supernatant were collected.Beclin-1 protein level and cell proliferaion rate were determined by western blotting and CCK-8 kit respectively.Results:With the treatment of LPS,Beclin-1 protein level was enhanced significantly,when compared with the control(P〈0.05).The cell proliferation rate decreased,and a signifant difference was found as well(P〈0.05).Conclusion:LPS can induce the activation of autophagy-related Beclin-1 protein,which may be invovled in the modulation of glia’ death.
