摘要
目的:探讨高压氧条件下微重力培养的高分子可降解聚乙醇酸支架-胰岛复合物移植后对糖尿病大鼠的治疗效果。方法:将胰岛细胞在体外静止培养(对照组)、支架微重力培养(实验组1)及高压氧条件下支架微重力培养(实验组2),5d后将胰岛移植到糖尿病大鼠体内。在移植后不同的时间点,分别对各组血糖及血清胰岛素含量等指标进行检测。结果:与对照组相比,实验组1和实验组2糖尿病大鼠的血糖浓度显著降低,血清胰岛素含量显著增高;且实验组2效果更为显著。电镜观察显示移植后,支架-胰岛复合物中胰岛形态良好,细胞间可见交织呈网状的细胞外基质。结论:高压氧条件下微重力培养可更加改善胰岛移植物的形态并使其更好地发挥生物学效能。
Objective:To investigate the efficacy of the transplantation of polyglycolic acid(PGA)-islet complex cultured in hyperbaric oxygen on diabetes model of Wistar rats.Methods: The islets were cultured in vitro in three conditions: the normal condition(control group);with PGA and simulated microgravity conditions(experimental group 1) and with PGA simulated microgravity and hyperbaric oxygen conditions(experimental group 2).Five days after cell culture,these islets were transplanted separately into diabetes wistar rats and the blood sugar concentrations,the insulin levels in plasma.Results: In experimental group 1 and experimental group 2,the blood sugar concentrations in plasma were significantly decreased and the levels of insulin were markly increased compared with that in control group.The results of scanning electron microscope indicated that there were many good-shape islets which could adhere to PGA under hyperbaric oxygen condition after transplantation.Conclusions: The islets cultured in hyperbaric oxygen conditions could improve the shape of islet-graft and therefore play a better role in the treatment of diabetes.
