产木聚糖酶和纤维素酶真菌的酶学性质分析 被引量:10

Screening of Cellulase and Xylanase-pruducing Fungi from Mangrove Soil and Characterization of the Enzymatic Reactions

摘要 红树林土壤微生物资源丰富,从深圳福田红树林土壤中分离了80株真菌,经过产酶发酵复筛,获得2株同时具有较高木聚糖酶和纤维素酶活的真菌。通过菌株形态观察和分子鉴定,确定菌株1为草酸青霉(Penicillium oxalicum),菌株2为曲霉属的Aspergillus piperis。菌株1的木聚糖酶和纤维素酶酶活分别为42.16 IU和3.05 IU,菌株2的木聚糖酶和纤维素酶酶活分别为39.01 IU和0.87 IU。同时,对其进行酶学性质研究表明,2菌株木聚糖酶的最适温度均为50℃;菌株1纤维素酶的最适温度是60℃,菌株2纤维素酶的最适温度是70℃,具有一定的耐热性。两菌株纤维素酶的最适pH均为3.0;菌株1木聚糖酶的最适pH为5.0,菌株2木聚糖酶的最适pH为4.0。 Mangrove ecological system has rich resources of microorganisms in the soil.Eighty strains of funfi were isolated from mangrove environment.Two strains of cellulase and xylanase-pruducing fungi were obtained after rescreening fermentation for enzyme production.The enzyme activities of cellulase and xylanase of strain No.1 is 42.16 IUand 3.05 IU respectively.The enzyme activities of cellulase and xylanase of strain No.2 is 39.01IU and 0.87 IU respectively.The optimum temperature of xylanase of two strains are all 50 ℃.The optimum temperature of cellulase of strain No.1 and No.2 are 60 ℃ and 70 ℃ respectively.The optimum pH of cellulase of two strains are all pH3.0.The optimum pH of xylanase of strain No.1 and No.2 are pH5.0 and pH4.0 respectively.Strain No.1 and No.2 were identified as Penicillium oxalicum and Aspergillus piperis respectively by morphologic observation and the result of ITS analysis.
