摘要
以超高压微射流均质(0 MPa、60 MPa、100 MPa、160 MPa)对低温花生粕蛋白预处理后再用Alcalase 2.4L酶解,发现60 MPa均质处理可提高蛋白回收率及水解度,且酶解产物的抗氧化能力指数(ORAC值)及DPPH.清除率也增加;酶解4 h后ORAC值达到最高(1943.6μmol Trolox equivalent/g peptide),DPPH.清除率也较高(71.90%),经凝胶色谱分析知产物中主要组分为1000~5000 Da的多肽(占总量的66.64%)。所得酶解物与玻璃珠的最佳吸附条件为:蛋白浓度2 mg/mL,吸附温度30℃,吸附时间7 h。解吸达到平衡时所得洗脱液的ORAC值及DPPH.清除率均有较好的活性保持率(分别为77.95%和79.35%)。
Microfluidization(0 MPa~160 MPa) was applied to treat peanut protein before proteolysis by Alcalase 2.4L.It was found that microfluidization treatment at 60MPa could enhance protein recovery and the degree of hydrolysis.Besides,the oxygen radical absorbance capacity(ORAC) value and DPPH· scavenging activity were also increased.The ORAC value reached the highest value(1943.6 μmol Trolox equivalent/ g Peptide) after hydrolysis for 4 h.Gel filtration chromatography analysis showed that the hydrolysate was dominated by 1000-5000 Da peptides(66.64%).The optimum conditions for the absorption of hydrolysates with glass beads were protein concentration 2mg/mL,absorption temperature 30°C and absorption time 7 h.The hydrolysate maintained 77.95% of its original ORAC value and 79.35% of its DPPH scavenging activity.
