摘要
研究吴茱萸碱对人肝癌细胞HepG-2的细胞周期阻滞作用及其机制.MTT法检测吴茱萸碱(Evodiamine)对人肝癌HepG-2细胞的细胞毒作用;流式细胞仪分析吴茱萸碱对HepG-2细胞周期的影响;免疫荧光染色法,经激光共聚焦显微镜(CLSM)观察检测吴茱萸碱对HepG-2细胞内微管蛋白α-tubulin聚合形态的影响.吴茱萸碱对人肝癌细胞HepG-2的IC50为19.62μmol/L;吴茱萸碱可将HepG-2细胞阻滞于G2/M期;免疫荧光,激光共聚焦显微镜观察可知,吴茱萸碱可显著改变微管蛋白聚合态发生,使细胞内微管呈束状分布,干扰纺锤体形成.实验表明吴茱萸碱通过影响微管蛋白的聚集状态,将HepG-2细胞周期阻滞于G2/M期.
To study the effect of cells cycle arrest and mechanism in human liver cancer HepG-2 which induced by evodiamine.The cytotoxic effect of evodiamine was determined by MTT assay.The influence of evodiamine on human liver cancer’s cell cycle distribution was examined by flow cytometry.Used immunofluorescence and observed by confocal laser microscopy to investigate the changes of tubulin.Treated HepG-2 cell line with different doses of evodiamine,for 24 hours,the IC50 was 19.62 μmol/L.Evodiamine could induce G2/M phase arrest.Under confocal laser microscopy,the polymeric form of tubulin was changed significantly,blocked the spindle poles formed.Evodiamine could induce cell cycle arrest in human liver cells line HepG-2,the mechanism had effect on polymeric form of tubulin and play the role of anticancer activity.
