摘要
对出发菌株进行紫外诱变以获得葡萄糖-1-磷酸产量高且遗传稳定性好的枯草芽孢杆菌菌株。以葡萄糖-1-磷酸的发酵产量为考察指标,采用HPLC-MS检测方法,对出发菌株Bacillus sublitis XH-13进行三轮紫外诱变,并对获得的高产菌株连续传代6次。结果表明,枯草芽孢杆菌突变菌株UV3-8的葡萄糖-1-磷酸产量最高,达到6.85 g/L,是出发菌株产量的1.54倍,且遗传稳定性好。菌株Bacillussubtilis XH-13_UV3-8的营养简单、葡萄糖-1-磷酸产量高且遗传稳定性好,具有产业化应用前景。
To obtain glucose-1-phosphate high-producing and genetic characteristics stable strain by treating Badllus sublitis XH-13 with UV. Badllus sublitis XH-13 was conducted three rounds of UV mutagenesis, and the mutant highest-producing glucose-1-phosphate detected by HPLC-MS was continuously passaged 6 genenation. Glucose-l-phosphate production of the mutant UV3-8 was the highest,6.85 mg/mL, 1.54 times of that of the parent strain, and its genetic characteristics was stable. As nutrition simple, glucose-1-phosphate production high and genetic characteristics stable, Bacillus subtilis XH-13_UV3-8 is an ideal strain to use in industrial production.
