双氢青蒿素对人结直肠癌裸鼠移植瘤的抑制作用及机制研究 被引量:7

Antitumor Effect and Mechanism of Dihydroartemisinin on Colorectal Cancer in Nude Mice with Transplanted Tumor of Human Colorectal Cancer

摘要 目的研究双氢青蒿素(Dihydroartemisinine,DHA)对人结直肠癌LoVo细胞裸鼠移植瘤的抑制作用及可能机制。方法将人结直肠癌LoVo细胞接种于Balb/c裸鼠皮下,建立结直肠癌模型。将造模成功裸鼠随机分为5组,即模型对照组,阳性对照组(5-Fu 20 mg/kg),DHA高、中、低剂量组(120,60,30 mg/kg)。观察各组动物肿瘤的重量及抑瘤率;采用免疫组化SABC法测定各组裸鼠肿瘤组织B-cell lymphoma-leukemia-2 gene(Bcl-2)和血管内皮生长因子(VEGF)的表达。结果 5-Fu组及DHA高、中剂量组的抑瘤率显著高于模型组(P<0.01或P<0.05);5-Fu组和DHA高剂量组Bcl-2的阳性表达率显著低于模型对照组(P<0.01或P<0.05);5-Fu组VEGF的阳性表达率显著低于模型对照组(P<0.05)。结论双氢青蒿素具有较强的抗结直肠癌活性,其作用机制可能是抑制抗凋亡基因Bcl-2的表达,从而刺激结直肠癌细胞凋亡。 Objective To observe the antitumor effect and mechanisms of dihydroartemisinin(DHA) on transplanted tumor of colorectal cancer in nude mice. Methods Colorectal cancer animal models were established by subcutaneous injection of colorectal cancer LoVo cells suspension into the specific-pathogen free BALB/c nude mice. Then the BALB/c tumor-bearing nude mice were randomized into 5 groups: model group, 5-FU group (intraperitoneal injec tion of 5-FU 20 mg/kg for 5 days) , and high-, middle-and low-dose DHA groups(gastric gavage of DHA in the dose of 120, 60 and 30 mg/kg respectively for 14 days). The tumor mass weight and tumor-inhibition rate were examined after treatment. And the expression levels of Bcl-2 and vascular endothelial growth factor(VEGF) were detected by SABC immunohistochemical assay. Results After treatment, the tumor-inhibition rate were increased in 5-FU group and high-and middle-dose DHA groups(P 〈 0.01 or P 〈 0.05 compared with those in the model group). The positive expression rate of bcl-2 in 5-FU group and high dose DHA was lower than that in the model group (P 〈 0.01 or P 〈 0.05), and the positive expression rate of VEGF in 5-FU group was also lower than that in the model group(P 〈 0.05). Conclusion DHA has been shown to exhibit certain antitumor activity against colorectal cancer in animal models. Its mechanism may be related to inducing the apoptosis of cancer cells by inhibiting the expression of Bcl-2.
