摘要
目的探讨4’-甲醚-黄芩素(4-MS)诱导人绒毛膜癌JAR细胞凋亡的分子机制。方法应用透射电镜、流式细胞术、实时定量PCR法和Western blotting方法,体外观察4-MS对JAR细胞的影响。结果电镜下可见用药后JAR细胞凋亡的形态学改变;流式细胞术显示,随着作用时间和药物剂量的增加,早期凋亡细胞和坏死细胞增加,明显高于对照组(P<0.01)。20、40 mg.L-1 4-MS作用48 h后c-myc mRNA的表达量为0.87±0.12和0.45±0.08;c-myc蛋白的表达量是0.37±0.02和0.25±0.02,均低于对照组(P<0.01)。结论 4-MS诱导JAR细胞凋亡,其机制与降低c-myc的表达有关。
Objective To investigate the molecular mechanism of the apoptosis of human choriocarcinoma JAR cell line induced by 4'-methery-scutellarein(4-MS). Methods The techniques of transmission electron microscopy, flow cytometry, real-time quantitative polymerase chain reaction(PCR) and Western blotting were used to study the effect of 4-MS on JAR cells in vitro. Results The morphological changes of apoptosis appeared in JAR cells after treatment with 4-MS. The results of flow cytometry showed that 4-MS induced early apoptosis and cellular necrosis in both con- centration- and time-dependent manners, much higher than those in the control group(P 〈 0.01 ). After the JAR cells were treated by 4-MS at the concentrations of 20 and 40 mg L-1 for 48 h, c-myc mRNA expression level were 0.87±0.12 and 0.45±0.08, and that of e-myc protein were 0.37±0.02 and 0.25±0.02, respectively. Both the expression levels of e-mye mRNA and c-mye protein were lower than those in the control group(P 〈 0.01). Conclusion 4-MS can induce JAR cell apoptosis, and its mechanism is related to the deerease of c-mye expression.
